Study of Hb Variants : Abnormality of Exon ¥± on the Xho ¥± Cleavage Site

Abstract
The human ¥â-globin gene has been known to be many DNA sequence variation, and some of these could be detectable as variants in the length of DNA fragments produced by a restriction endonuclease digestion. Accordingly, I investigated the genetic variation on human ¥â-globin coding region. At first, I found the abnormal ¥â-chain of hemoglobin in Korean by aga gel, urea-starch gel and Supersepraphore ¥² polyacetate membrane electrophoresis. Polymerase chain reaction(PCR) for detection of Exon ¥°-¥±, ¥±, and ¥² in ¥â-globin gene was performed on normal and abnormal ¥â-globin specimens, which were found at Soonchunhyang University Chunan Hospital by agar gel electrophoresis. Comparison study was made among the DNA fragments obtained from the restriction endonudease digests of PCR products. From the analysis of direct restriction enzyme digestion, I revealed a genetic variation in a Xho¥± recognition site(-GGATCT-) about gene sequences coding ¥â globin.
